With artificial reproduction techniques, semen samples must be first processed before they can be used for insemination. Dr. K.D. Nayar, Gynaecologist & Obstetrician, HOD, Akansha IVF Centre, Mata Chanan Devi Hospital, New Delhi, discusses the components and contributions of semen, normal semen sample, sperm motility and sperm morphology evaluation. The various sperm preparation techniques (simple wash, swim up technique, density gradient, swim down, glass wool filtration and magnetic activated cell sorting) are discussed in detail.
Dr. Nayar also explains the technique of retrograde ejaculation, preparation of epididymal and testicular spermatozoa, selection methods to isolate the best sperm, sperm selection by intra cytoplasmic morphologically selected sperm injection (IMSI) and motility sperm organelle morphology examination (MSOME) and the factors affecting success rate.
